چکیده
Abstract Lyme disease (LD) which is caused by the spirochete Borrelia burgdorferi (Bb) poses a global health threat. Bb infection induces type I IFN response that associated with arthritis and other symptoms. However, ligands, PAMPs, signaling pathways production are poorly characterized. Stimulator of Interferon Genes (STING) pathway an intracellular surveillance detects c-di-nucleotides (CDNs) produced pathogens through STING production. produces CDNs hence it has potential to activate pathway. In this study, role c-di-AMP in elicitation was elucidated. The RAW cell line infected activation measured detecting downstream proteins (pSTING, pTBK1, pIRF3 & pNF-κB), proinflammatory cytokines (IL6, TNF-α) Type (IFN-β) using ELISA, western blotting, immunostaining, Luciferase Reporter assay. macrophage results mediated response. induction level dependent on as mutant strain producing higher induced cytokine Interestingly, released into BSK-II medium during culture could also stimulate protein KO showed reduction IFN-β. conclusion, our show Bb-produced major PAMP triggers interferon

STING-Dependent Recognition of Cyclic di-AMP Mediates Type I Interferon Responses during Chlamydia trachomatis Infection
UNLABELLED STING (stimulator of interferon [IFN] genes) initiates type I IFN responses in mammalian cells through the detection of microbial nucleic acids. The membrane-bound obligate intracellular bacterium Chlamydia trachomatis induces a STING-dependent type I IFN response in infected cells, yet the IFN-inducing ligand remains unknown. In this report, we provide evidence that Chlamydia synthe...
متن کاملI-43: Expression Profile of Macrophage Migration Inhibitory Factor (MIF) Signaling Pathway as A Potentional Biomarker in Pathophysiology of Endometriosis
Background MIF via its receptor, CD74, initiates a signaling cascade that leads to proliferation and survival of cells. Also, MIF binding to CD74 activates p38 signaling pathways that lead to positive effect on the expression of COX-2. The aim of this study was to evaluate the gene expression profile of MIF, CD74 and COX-2 in normal, ectopic and eutopic endometrium during menstrual cycle. The e...
متن کاملMPYS is required for IFN response factor 3 activation and type I IFN production in the response of cultured phagocytes to bacterial second messengers cyclic-di-AMP and cyclic-di-GMP.
Cyclic-di-GMP and cyclic-di-AMP are second messengers produced by bacteria and influence bacterial cell survival, differentiation, colonization, biofilm formation, virulence, and bacteria-host interactions. In this study, we show that in both RAW264.7 macrophage cells and primary bone marrow-derived macrophages, the production of IFN-β and IL-6, but not TNF, in response to cyclic-di-AMP and cyc...
متن کاملType III interferon (IFN) induces a type I IFN-like response in a restricted subset of cells through signaling pathways involving both the Jak-STAT pathway and the mitogen-activated protein kinases.
Type III interferon (IFN) is a novel member of the interferon family. Type III IFN utilizes a receptor complex different from that of type I IFN, but both types of IFN induce STAT1, STAT2, and STAT3 activation. Here we describe a detailed comparison of signal transduction initiated by type I and type III IFN. Gene expression array analysis showed that IFN types I and III induced a similar subse...
متن کاملPanax ginseng induces human Type I collagen synthesis through activation of Smad signaling.
Skin aging appears to be principally related to a decrease in levels of Type I collagen, the primary component of the dermal layer of skin. It is important to introduce an efficient agent for effective management of skin aging; this agent should have the fewest possible side effects and the greatest wrinkle-reducing effect. In the course of screening collagen production-promoting agents, we obt...
